CpG-2722 is a synthetic CpG-oligodeoxynucleotide (CpG-ODN) that functions as a Toll-like receptor 9 (TLR9) agonist. Developed for cancer immunotherapy, it stimulates the innate immune system by mimicking bacterial DNA motifs, which leads to the maturation of dendritic cells, the production of pro-inflammatory cytokines, and the recruitment of immune cells to the tumor microenvironment. Preclinical research, particularly in head and neck cancer models, has demonstrated that CpG-2722 also induces the exposure of phosphatidylserine (PS) on the surface of cancer cells. This unique mechanism can be leveraged in combination therapies with PS-targeting agents to enhance antitumor efficacy through immunogenic cell death and improved drug targeting.
