Overview
CpG-28 oligodeoxynucleotide is a synthetic oligodeoxynucleotide containing unmethylated CpG motifs derived from bacterial DNA, designed to stimulate the immune system. It acts as an agonist of Toll-like receptor 9 (TLR9), leading to activation of both innate and adaptive immunity, particularly driving a Th1-type immune response. In clinical studies, it has been investigated primarily for use in cancer immunotherapy, including recurrent glioblastoma and neoplastic meningitis. The drug is administered either intratumorally or via subcutaneous/intrathecal routes and has shown tolerability with mainly transient side effects such as fever and lymphopenia[1][2][3]. The developer is Oligovax.
