Overview

CPG-52364 is a small molecule antagonist of toll-like receptors TLR7, TLR8, and TLR9. It was developed by Coley Pharmaceutical Group (later acquired by Pfizer) as an orally administered immunosuppressive agent for the treatment of immune-mediated diseases, including systemic lupus erythematosus. The compound selectively inhibits signaling through TLR7, TLR8, and TLR9—receptors that recognize certain nucleic acids such as CpG DNA and single-stranded RNA—thereby reducing undesirable immune activity. Clinical development reached phase 1 trials for systemic lupus erythematosus but was discontinued before further advancement[2][3][4][5].
