CpG-6tdGO is a preclinical bifunctional synthetic CpG oligodeoxynucleotide engineered to deliver multiple 6-thio-2′-deoxyguanosine nucleosides into scavenger-receptor-positive, Toll-like receptor 9-positive myeloid leukemia cells. Following cellular uptake and intracellular release, the 6tdG payload is used by telomerase reverse transcriptase and incorporated into telomeric chromatin, causing telomere DNA damage and rapid cytotoxicity in telomerase-positive acute myeloid leukemia cells. The CpG domain also activates Toll-like receptor 9, and treatment-associated tumor cell death can stimulate STING-dependent innate immunity and CD8-positive T-cell-mediated antitumor responses. The program was developed at City of Hope and has shown preclinical activity in primary human AML xenografts and syngeneic AML models while sparing hematopoietic stem cells and activated T cells in reported studies.
