Overview

CPG 7909 + dacarbazine is a **combination therapy** investigated primarily in metastatic melanoma. **CPG 7909** is a synthetic CpG oligodeoxynucleotide acting as a **Toll-like receptor 9 (TLR9) agonist**, which stimulates plasmacytoid dendritic cells and B cells, activating both innate and adaptive immunity, and resulting in secretion of proinflammatory cytokines and development of a Th1 cytotoxic environment[1][2][5][9]. **Dacarbazine** is an alkylating agent used as a chemotherapy drug. The combination aims to enhance anti-tumor immune responses in cancers, particularly melanoma, by combining immunomodulation (via CPG 7909) with the cytotoxic activity of dacarbazine[1][5][9]. Clinical trials indicate that this combination is generally well-tolerated, with manageable side effects (mainly transient injection site reactions, fever, arthralgias) but has not shown significant clinical benefit over dacarbazine alone in metastatic melanoma[1][5][9].
