**CpG-B oligodeoxynucleotides** are a heterogeneous, target-defined family of synthetic single-stranded oligodeoxynucleotides rather than one unique active pharmaceutical ingredient. They generally contain unmethylated cytosine-phosphate-guanine motifs on a phosphorothioate backbone and act as **Toll-like receptor 9 agonists**. Endosomal TLR9 activation primarily stimulates B cells and antigen-presenting cells, promoting NF-kB- and MAPK-associated inflammatory signaling, B-cell activation, cytokine production, and adaptive immune priming; CpG-B agents generally induce less type I interferon than CpG-A agents. Individual CpG-B sequences and formulations have been investigated as cancer immunotherapies and vaccine adjuvants, but this class name does not identify a single clinical product, sponsor, formulation, or INN.
