Overview

CpG oligodeoxynucleotide 1668 is a **class B synthetic oligonucleotide containing unmethylated cytosine-guanine (CpG) motifs with phosphorothioate backbone** that acts as a Toll-like receptor 9 (TLR9) agonist, specifically stimulating murine TLR9[4][1]. This compound is a 20-mer DNA sequence (5’-tccatgacgttcctgatgct-3’) engineered for nuclease resistance[4]. ODN 1668 induces strong **immunostimulatory effects**, leading to enhanced Th1-mediated CTL activation, pro- and anti-inflammatory cytokine production (IL-1Ra, IL-1α, IL-1β, IL-6, TNFα, IL-10), and nitric oxide release from macrophages[2][1]. Its primary biomedical use is as an experimental **immunomodulatory adjuvant** in preclinical models of cancer immunotherapy, especially murine models, where it enhances anti-tumor immunity, mainly via Th1 responses and CTL activation[1]. Unlike class A CpG ODNs, ODN 1668 does not directly activate NK cells but is a potent Th1 activator[1].
