CpG oligoDNA, also known as CpG oligodeoxynucleotides (CpG ODNs), is a class of synthetic, short, single-stranded DNA molecules containing unmethylated cytosine-guanine (CpG) dinucleotide motifs. These motifs mimic the DNA found in bacteria and viruses, serving as potent agonists of Toll-like Receptor 9 (TLR9). Activation of TLR9 by CpG ODNs triggers innate and adaptive immune responses, including the activation of B cells, plasmacytoid dendritic cells, NK cells, and cytotoxic T lymphocytes. They are primarily investigated as vaccine adjuvants and anticancer immunotherapies for various indications, including glioblastoma, mesothelioma, and infectious diseases. While several specific CpG ODNs (e.g., CpG 1018, CpG 7909) have been developed by different companies, CpG oligoDNA itself refers to the broader class of these nucleic acid therapeutics.
