CPG10101 + ribavirin is an investigational combination therapy studied for the treatment of chronic hepatitis C virus (HCV) infection, particularly in patients who are refractory or have relapsed following previous therapy. **CPG10101** (also known as ACTILON) is a synthetic oligodeoxynucleotide that acts as a Toll-like receptor 9 (TLR9) agonist, stimulating dendritic cell-mediated and B cell-mediated immune responses[4][5]. **Ribavirin** is a broad-spectrum antiviral nucleoside analog that inhibits viral RNA synthesis. The combination was explored for its synergy in enhancing antiviral effects, particularly when combined with pegylated interferon-alpha. Clinical studies demonstrated dose-dependent immune activation, increased early virologic response (EVR), and improved reduction in HCV RNA levels compared to standard regimens. CPG10101 was generally well tolerated, with most adverse events being mild or moderate, consisting primarily of flu-like symptoms and injection site reactions[4][5]. Development of CPG10101 for HCV has been suspended and is no longer pursued for this indication[1][4][5].
