CPI-006 + ciforadenant + pembrolizumab is an investigational combination therapy composed of three distinct agents with complementary mechanisms used in oncology clinical trials. CPI-006 is a humanized monoclonal antibody targeting CD73, designed to inhibit adenosine production and thereby modulate the immunosuppressive tumor microenvironment. Ciforadenant is a small molecule antagonist of the adenosine A2A receptor, reducing adenosine-mediated immunosuppression and promoting T cell activity. Pembrolizumab is a humanized monoclonal antibody (IgG4) targeting programmed cell death protein 1 (PD-1) on T cells, blocking the interaction with its ligands PD-L1 and PD-L2 to restore and enhance anti-tumor immune responses[1][2][3][4]. This combination is developed for cancers, particularly in immunotherapy-resistant, high adenosine signature, or checkpoint-inhibitor refractory tumors.
