CPI-006 is a potent, investigational humanized monoclonal antibody that targets CD73, a cell-surface ectonucleotidase involved in the production of immunosuppressive adenosine within the tumor microenvironment. By inhibiting CD73, CPI-006 blocks adenosine production and modulates immune responses by activating lymphocytes and inducing B cell activation and trafficking[1][3][6]. Pembrolizumab is an approved monoclonal antibody targeting PD1 (programmed cell death protein 1), used as an immune checkpoint inhibitor in various cancers. The combination of CPI-006 with pembrolizumab is being investigated for synergistic immunomodulatory effects to enhance antitumor immunity in patients with advanced solid tumors[3][7]. This combination has been evaluated in phase 1/1b clinical trials for multiple cancer types including non-small cell lung cancer, renal cell carcinoma, colorectal cancer, triple-negative breast cancer, cervical cancer, ovarian cancer, pancreatic cancer, endometrial cancer, sarcoma, squamous cell carcinoma of the head and neck, bladder cancer, metastatic castration-resistant prostate cancer and non-Hodgkin lymphoma[5][8].
