Drug intelligence / Profile preview
CPL-1 (Cpl-1) is a recombinant bacteriophage-derived endolysin (specifically, a peptidoglycan hydrolase or muramidase) originally encoded by the pneumococcal bacteriophage Cp-1. It specifically targets and hydrolyzes the cell wall peptidoglycan of *Streptococcus pneumoniae* (pneumococcus) by cleaving the glycosidic bond between N-acetylmuramic acid and N-acetylglucosamine, leading to rapid osmotic lysis and bacterial death. CPL-1 has demonstrated high bactericidal efficacy and specificity against both antibiotic-susceptible and multidrug-resistant pneumococcal strains in various preclinical models of pneumonia, bacteremia, meningitis, otitis media, and endocarditis. It is being investigated as a potential therapeutic agent (enzybiotic) for pneumococcal infections, either as a purified recombinant protein or via host-expressed synthetic mRNA delivery.
