Overview

CPL423 is a potent, small-molecule multi-kinase inhibitor developed by Celon Pharma. It targets the TAM receptor tyrosine kinase family—comprising TYRO3, AXL, and MERTK—alongside the FMS-like tyrosine kinase 3 (FLT3). These kinases are frequently dysregulated in various cancers, contributing to tumor survival, metastasis, and resistance to conventional therapies. By inhibiting the TAM family, CPL423 not only blocks direct oncogenic signaling but also modulates the tumor microenvironment to overcome immune evasion, notably by enhancing tumor phagocytic capacity. It is primarily being developed for the treatment of acute myeloid leukemia (AML) and various solid tumors where TAM or FLT3 dysregulation is a driver of disease progression.
