CPP-MAGE-A3 is a **fusion protein composed of the tumor antigen MAGE-A3 (Melanoma Antigen Gene-A3) covalently linked to a cell-penetrating peptide (CPP)**, designed to enhance the intracellular delivery of the antigen to dendritic cells for therapeutic cancer immunization. The addition of a CPP (such as YARKARRQARR) increases the cytosolic uptake of MAGE-A3, improving the antigen presentation on HLA class I molecules and thereby potentially generating a stronger cytotoxic T lymphocyte (CTL) response against MAGE-A3-expressing tumor cells. This strategy targets immune activation against cancers expressing MAGE-A3, such as melanoma and pancreatic cancer. Preclinical and ex vivo studies have shown enhanced dendritic cell uptake, increased IFN-γ secretion, and improved tumor cell lysis compared to MAGE-A3 alone. This construct is used primarily in experimental dendritic cell-based immunotherapy approaches[1][2].
