Overview

CPP-P1 is a neuroprotective chimeric peptide composed of **peptain-1**—a 10-amino acid sequence (DRFSVNLDVK) derived from the chaperone domain of **alphaB-crystallin (CRYAB)**—conjugated to a **cell-penetrating peptide (CPP)** to facilitate intracellular delivery. It is primarily being investigated for the treatment of **glaucoma** and **ocular hypertension**. CPP-P1 exerts its neuroprotective effects by activating the **cAMP response element-binding protein (CREB)** signaling pathway, which promotes the survival of **retinal ganglion cells (RGCs)** and preserves the structural integrity of retinal layers, including the inner nuclear and plexiform layers. Additionally, treatment with CPP-P1 has been shown to upregulate synaptic and growth-associated markers such as **synapsin-1 (SYN1)** and **growth-associated protein 43 (GAP43)**, suggesting a role in maintaining neuronal connectivity and regenerative potential under conditions of elevated intraocular pressure.
