Overview

CPS (Chemoprophylaxis and Sporozoites) immunization is an experimental whole-parasite malaria vaccine strategy developed by Radboud University Medical Center. The approach involves immunizing healthy volunteers with live, infectious Plasmodium falciparum sporozoites (typically the NF54 strain) delivered through the bites of infected mosquitoes while the subjects are under chloroquine prophylaxis. Chloroquine acts as a blood-stage schizonticide, preventing clinical malaria by killing parasites as they emerge from the liver, while allowing liver-stage development to proceed. This exposure to pre-erythrocytic and early erythrocytic antigens induces potent, sterile protective immunity against subsequent homologous sporozoite challenge. Clinical trials have demonstrated that this approach can induce sterile protection in over 90% of volunteers. The primary biological targets of the induced immune response include the circumsporozoite protein (CSP) and liver stage antigen 1 (LSA1).
