Overview

CPS (chemoprophylaxis and sporozoite) immunization is an experimental whole-parasite malaria vaccine strategy developed by Radboud University Medical Center. This specific approach utilizes the Plasmodium falciparum NF135.C10 strain, a Cambodian isolate selected for its ability to produce high numbers of large liver-stage schizonts. During the immunization process, healthy volunteers are exposed to the bites of Anopheles mosquitoes infected with NF135.C10 sporozoites while concurrently receiving antimalarial chemoprophylaxis (typically mefloquine or artemether/lumefantrine). This allows the parasites to infect the liver and induce a broad repertoire of protective immune responses against pre-erythrocytic stages, while the chemoprophylaxis prevents the development of symptomatic blood-stage malaria. Clinical trials, such as NCT03813108, have evaluated the safety, tolerability, and protective efficacy of this regimen against both homologous and heterologous parasite challenges.
