CPT-732 is a preclinical-stage molecular glue degrader (MGD) developed by Captor Therapeutics that specifically targets NIMA-related kinase 7 (NEK7). NEK7 is a critical regulator of the NLRP3 inflammasome, a multi-protein complex that triggers the release of pro-inflammatory cytokines such as IL-1β and IL-18 in response to various danger signals. By inducing the targeted proteasomal degradation of NEK7, CPT-732 aims to inhibit the assembly and activation of the NLRP3 inflammasome, thereby suppressing the downstream inflammatory cascade. This mechanism of action is being explored for the treatment of a wide range of autoimmune and neuroinflammatory conditions, including rheumatoid arthritis, psoriasis, multiple sclerosis, amyotrophic lateral sclerosis (ALS), and Huntington's disease.
