Overview

CPT113 is a proprietary Topoisomerase I (TOPO1) inhibitor payload and linker-payload technology developed for use in antibody-drug conjugates (ADCs). It serves as the cytotoxic warhead that, upon release within target cancer cells, inhibits DNA Topoisomerase I, leading to lethal DNA strand breaks and apoptosis. CPT113 is a key component of the CrossConju™ conjugation platform, which is designed to enable high drug-to-antibody ratios (DAR) and enhanced stability. It is incorporated into several ADC candidates, including DXC1002 (targeting TROP2) and DXC006 (targeting CD56), as well as programs targeting PTK7, SEZ6, and MUC16. The technology was developed by researchers at Duality Biologics.
