Overview

CPV-154 is a thiosulfinate derivative and a small molecule inhibitor of thioredoxin reductase 1 (TrxR1). Developed by researchers at the University of Puerto Rico, CPV-154 was designed as part of a library of cyclic disulfides and thiosulfinates targeting the redox-active sites of human thioredoxin reductase. The thioredoxin system is overexpressed in various cancers, promoting cell proliferation and survival while inhibiting apoptosis. In preclinical screenings, CPV-154 demonstrated significant cytotoxic activity against a panel of breast cancer cell lines, including MCF-7, ZR-75-1, BT-20, MDA-MB-468, SkBr-3, BT-474, and MDA-MB-231, by inhibiting cell growth and inducing oxidative stress.
