Drug intelligence / Profile preview
CPX-351 (Vyxeos) is a liposomal formulation co-encapsulating the anthracycline daunorubicin and the pyrimidine analogue cytarabine in a fixed 1:5 molar ratio. This specific ratio has been shown in preclinical models to maximize synergistic antileukemic activity. The liposomal delivery system (CombiPlex platform) enhances the pharmacokinetics of the drugs, allowing for prolonged exposure and preferential uptake by leukemic cells in the bone marrow. Daunorubicin acts by intercalating into DNA and inhibiting topoisomerase II, while cytarabine acts as an antimetabolite that inhibits DNA polymerase and incorporates into DNA, leading to chain termination. CPX-351 is FDA-approved for the treatment of adults with newly-diagnosed therapy-related acute myeloid leukemia (t-AML) or AML with myelodysplasia-related changes (AML-MRC).
