Drug intelligence / Profile preview
CPX-351 (Vyxeos) is a liposomal encapsulation of the antineoplastic agents cytarabine and daunorubicin, maintained at a fixed 5:1 molar ratio. This formulation is designed to deliver the two drugs synergistically to leukemia cells while limiting systemic exposure and toxicity. Cytarabine, a pyrimidine analog, inhibits DNA polymerase and DNA synthesis, while daunorubicin, an anthracycline, intercalates into DNA and inhibits topoisomerase II. CPX-351 is FDA-approved for the treatment of adults and pediatric patients (1 year and older) with newly-diagnosed therapy-related acute myeloid leukemia (t-AML) or AML with myelodysplasia-related changes (AML-MRC). It is also being investigated for use in higher-risk myelodysplastic syndromes (MDS).
