Drug intelligence / Profile preview
CPX-351 (Vyxeos) is a liposomal encapsulation of cytarabine and daunorubicin in a fixed 5:1 molar ratio. This formulation is designed to optimize the synergistic ratio of the two drugs while protecting them from degradation and enhancing delivery to the bone marrow. It is primarily used for the treatment of newly-diagnosed therapy-related acute myeloid leukemia (t-AML) or AML with myelodysplasia-related changes (AML-MRC). The mechanism involves cytarabine's inhibition of DNA polymerase and daunorubicin's intercalation into DNA and inhibition of topoisomerase II. The liposomal delivery system allows for prolonged exposure and preferential uptake by leukemia cells in the bone marrow.
