Overview

**CPX-351 + enasidenib** is a combination regimen of CPX-351—a liposomal encapsulation of cytarabine and daunorubicin at a fixed 5:1 molar ratio—and enasidenib, an oral inhibitor of isocitrate dehydrogenase 2 (IDH2). CPX-351 is designed to optimize the delivery and synergy of cytarabine (a nucleoside analog inhibiting DNA synthesis) and daunorubicin (an anthracycline inducing DNA damage), primarily for the treatment of acute myeloid leukemia (AML) with adverse or secondary features. Enasidenib targets mutant IDH2 enzymes, restoring normal cellular differentiation in IDH2-mutant AML. The combination is being evaluated in clinical trials for relapsed/refractory AML, especially in those with IDH2 mutations, as the dual approach may increase efficacy compared to either therapy alone[1][3].
