CPX-351 + venetoclax is a combination regimen consisting of CPX-351 (a dual-drug liposomal formulation containing cytarabine and daunorubicin in a fixed 5:1 molar ratio) and venetoclax (a selective small molecule inhibitor of the anti-apoptotic protein BCL-2). CPX-351 acts as an antineoplastic agent by inhibiting DNA synthesis and promoting DNA damage, while venetoclax promotes apoptosis by inhibiting BCL-2. The combination is being investigated in acute myeloid leukemia (AML), including relapsed/refractory and newly diagnosed patients unfit for intensive chemotherapy, as well as pediatric patients. Preclinical data indicate synergistic pro-apoptotic effects. Combination therapy has demonstrated promising response rates and a safety profile consistent with those of the individual agents[1][3][5][6].
