**CPX-571** is Celator Pharmaceuticals' preclinical liposomal fixed-dose combination of irinotecan and cisplatin, co-encapsulated at a 7:1 molar ratio using the CombiPlex drug-ratio delivery platform. The formulation was designed to maintain a synergistic irinotecan-to-cisplatin exposure ratio after intravenous administration. Irinotecan inhibits DNA topoisomerase I following conversion to its active metabolite SN-38, while cisplatin forms cytotoxic DNA crosslinks. CPX-571 showed superior antitumor activity versus the corresponding free-drug combination in preclinical xenograft models and was proposed for development in small cell lung cancer.
