CR-001

CR-001 is a tetravalent bispecific antibody that targets both the programmed cell death protein 1 (PD-1) receptor and vascular endothelial growth factor (VEGF). It was originally developed by Crescent Biopharma, a spinout from Paragon Therapeutics, and is now part of GlycoMimetics following an acquisition. The drug is designed to provide cooperative binding pharmacology for dual blockade of PD-1 and VEGF pathways, aiming to enhance anti-tumor immune responses while inhibiting angiogenesis in solid tumors. This mechanism mirrors the format and pharmacology of ivonescimab, which has shown superior efficacy compared to pembrolizumab in non-small cell lung cancer trials. CR-001 is currently in preclinical development with plans for an IND submission in late 2025 and proof-of-concept clinical data expected in 2026[2][4][5][10].
