CR-0305 is a preclinical-stage small molecule nitric oxide (NO) donor being developed by Coeurative for the treatment of refractory angina and coronary artery disease. Described as having a 'Breakthrough Dual-Action Mechanism,' the drug promotes vascular smooth muscle relaxation and coronary vasodilation. In ex vivo human coronary artery studies, CR-0305 demonstrated 23-fold greater vasodilatory potency than isosorbide mononitrate (ISMN), with an IC50 of 3.0 µM compared to 67.8 µM for ISMN. The program is currently in preclinical development, with a first-in-human (FIH) study planned for 2027.
