Overview

CR-1 is a near-infrared-II (NIR-II) croconaine dye that serves as a pH-sensitive theranostic agent. In the context of oncology, particularly hepatocellular carcinoma (HCC), CR-1 is utilized as the core imaging and therapeutic component of smart nanoprobes, such as the self-assembling CR-PEG-GBP conjugate. This molecule is designed to respond to the acidic tumor microenvironment (pH ~6.5), where it undergoes in situ self-assembly from small nanoprobes into larger nanoparticles. This aggregation enhances its tumor accumulation and retention time while facilitating dual-modal imaging (NIR-II fluorescence and photoacoustic imaging) and dual-modal therapy, including photothermal therapy (PTT) and sonodynamic therapy (SDT). By generating heat and reactive oxygen species (ROS) upon activation by light or ultrasound, CR-1 provides a targeted approach to tumor ablation with high detection sensitivity and deep tissue penetration.
