Overview

CR-2 peptide (mutant) is a synthetic peptide variant derived from the CR-2 domain of the GATAD2A protein. It was developed as a research tool to investigate the protein-protein interaction between GATAD2A and CHD4 within the MBD2-NuRD (Methyl-CpG-binding domain protein 2 - Nucleosome Remodeling and Deacetylase) complex. The MBD2-NuRD complex is a critical epigenetic repressor of fetal hemoglobin (HbF) in adult erythroid cells. While the wild-type CR-2 peptide is designed to disrupt the GATAD2A-CHD4 interaction and thereby induce HbF expression, the mutant version contains specific amino acid substitutions at the binding interface that abolish its affinity for CHD4. In preclinical studies, this mutant peptide serves as a negative control to demonstrate that the induction of HbF is specifically dependent on the disruption of the GATAD2A-CHD4 interface, supporting the development of small-molecule peptidomimetics for the treatment of sickle cell disease and beta-thalassemia.
