Overview

CR2-Crry is a recombinant fusion protein designed for targeted complement inhibition, primarily utilized in preclinical research. It consists of the C3-binding domain of mouse complement receptor 2 (CR2/CD21) fused to the mouse complement regulatory protein Crry. The CR2 domain acts as a targeting moiety, localizing the construct to sites of complement activation where C3 fragments (such as C3b, iC3b, and C3d) are deposited. The Crry domain provides broad-spectrum inhibition of the classical, lectin, and alternative complement pathways by acting as a decay-accelerating factor and a cofactor for Factor I, thereby preventing the formation and activity of C3 convertases. This site-specific approach allows for potent local anti-inflammatory effects while sparing systemic complement activity, which helps maintain host defense against infections. CR2-Crry has been extensively studied in various mouse models, demonstrating therapeutic potential in conditions such as ischemia-reperfusion injury, lupus-like renal disease, atherosclerosis, and traumatic brain injury.
