CR3294 is an investigational small molecule benzamidine derivative developed by Rottapharm S.p.A. for the prevention and treatment of chemotherapy-induced oral and gastrointestinal mucositis. Its mechanism of action involves the inhibition of reactive oxygen species (ROS) production and the release of pro-inflammatory cytokines from stimulated leukocytes, thereby mitigating the inflammatory cascade that leads to mucosal injury. In clinical development, CR3294 has been evaluated in Phase II multicenter, randomized, double-blind, placebo-controlled trials involving adult patients with hematologic malignancies undergoing high-dose chemotherapy (such as BEAM or high-dose melphalan) and autologous hematopoietic stem cell transplantation (HSCT).
