Overview

CRA-028129 is a small molecule inhibitor of cathepsin S, originally developed by Celera Genomics in collaboration with Bayer Schering Pharma. Cathepsin S is a lysosomal cysteine protease involved in MHC class II-mediated antigen presentation; its inhibition is intended to modulate the immune response in autoimmune and inflammatory conditions. CRA-028129 was primarily investigated as an oral therapy for psoriasis. It entered Phase I clinical trials in 2005 to evaluate safety and tolerability in healthy volunteers. However, development appears to have stalled, with no significant updates reported since approximately 2010.
