Overview

CRAd-S-pk7 is a conditionally replicative oncolytic adenoviral vector engineered for tumor specificity and enhanced infectivity. It utilizes the survivin promoter (S), which is highly active in cancer cells but minimally expressed in normal tissue, to drive selective viral replication within tumors. The pk7 modification of the adenoviral fiber protein increases the virus's ability to infect tumor cells. In clinical applications, CRAd-S-pk7 is delivered using allogeneic neural stem cells (NSCs) that home to tumors and protect the virus from immune clearance while facilitating targeted delivery. Upon administration, the NSCs release CRAd-S-pk7 at tumor sites; the virus then replicates selectively in cancer cells, causing direct lysis and stimulating an anti-tumor immune response through exposure of tumor antigens. This approach has been tested primarily for recurrent high-grade gliomas and investigated preclinically for ovarian cancer and endometriosis[2][3][4][5][6].
