Overview

CRB-556 is a peripherally restricted Cannabinoid Receptor 1 (CB1) inverse agonist currently in preclinical development by Corbus Pharmaceuticals. It was licensed as part of a library of over 600 cannabinoid compounds from Jenrin Discovery. Unlike first-generation CB1 inverse agonists like rimonabant, which were associated with significant neuropsychiatric side effects due to central nervous system (CNS) penetration, CRB-556 is designed to target CB1 receptors in peripheral tissues such as the liver, adipose tissue, and kidneys. This peripheral restriction aims to treat metabolic and fibrotic diseases, including obesity, non-alcoholic steatohepatitis (NASH), and chronic kidney disease, while minimizing CNS-related safety concerns. Preclinical studies have demonstrated its potential to improve metabolic parameters and reduce fibrosis in animal models.
