Drug intelligence / Profile preview
CRB2117 is a novel bispecific T-cell engager (TCE) targeting glypican-3 (GPC3) and CD3, being developed for the treatment of GPC3-positive hepatocellular carcinoma (HCC) [1747eP]. It is designed in a 2:1 format featuring bivalent, high-affinity GPC3-binding arms and an affinity-optimized CD3-binding arm [1747eP]. The CD3 arm is sterically hindered by the GPC3 arms to reduce systemic CD3 activation and mitigate cytokine release syndrome (CRS) risk, while the bivalent GPC3 arms enhance tumor avidity [1747eP]. In preclinical models, CRB2117 demonstrated potent T-cell-dependent cellular cytotoxicity (TDCC) and robust antitumor efficacy with a favorable safety profile and pharmacokinetic properties [1747eP].
