CRD-2015

CRD-2015 is a novel, potent, and highly selective small molecule inhibitor of calcium/calmodulin-dependent protein kinase II (CaMKII), currently under development by Cardurion Pharmaceuticals. With a reported IC50 of 0.62 nM, the compound works by reducing the phosphorylation of key CaMKII substrates, specifically targeting phospholamban (PLN) and ryanodine receptor 2 (RyR2) at their CaMKII-specific sites. This mechanism is designed to address the underlying pathophysiology of various cardiovascular diseases, including heart failure and arrhythmias such as catecholaminergic polymorphic ventricular tachycardia (CPVT) and atrial fibrillation. Preclinical studies have demonstrated that oral administration of CRD-2015 provides dose-dependent efficacy and significantly prolongs survival in animal models of severe heart failure, exhibiting a clear pharmacokinetic/pharmacodynamic relationship.
