Overview

CRD1400 is a potent, small molecule inhibitor of acetyl-CoA synthetase short-chain family member 2 (ACSS2) developed by Curadev Pharma. It targets the metabolic pathway where acetate is converted into acetyl-CoA, a critical process for tumor cell survival and lipid synthesis, especially in hypoxic and nutrient-depleted microenvironments. By blocking ACSS2, CRD1400 disrupts the energy supply and building blocks for acetate-dependent tumors, such as gliomas and triple-negative breast cancer, and reduces lipid accumulation in metabolic conditions like hepatic steatosis. It has shown the ability to sensitize cancer cells to chemotherapy and is currently in preclinical development.
