CRD1601 is a potent and selective small molecule inhibitor of Hematopoietic Progenitor Kinase 1 (HPK1), currently in preclinical development by Curadev Pharma for the treatment of various cancers. HPK1 is a member of the MAP4K family that functions as a negative regulator of T-cell receptor (TCR) and B-cell receptor (BCR) signaling. By inhibiting HPK1, CRD1601 prevents the kinase from inactivating immune cells, thereby enhancing anti-tumor immune responses through immune checkpoint modulation. High expression of HPK1 has been correlated with reduced patient survival in several cancer types, making it a significant target for cancer immunotherapy. The program aims to restore inflammatory immune responses by blocking HPK1-driven phosphorylation that down-regulates signaling in T and B cells.
