Overview

CRD3874 is a first-in-class, systemically administered small molecule allosteric agonist of the stimulator of interferon genes (STING) protein. Unlike traditional STING agonists that bind competitively to the canonical site, CRD3874 binds allosterically and activates the canonical STING pathway while simultaneously blocking STING’s proton channel activity. This dual action promotes robust anti-tumor immune responses by inducing type I interferons and pro-inflammatory cytokines without triggering excessive autophagy or pyroptosis, which are associated with toxicity in other STING agonists. Preclinical studies have demonstrated potent anti-cancer activity and favorable safety profiles in animal models. The drug is currently being evaluated in phase 1 clinical trials for advanced sarcoma and Merkel cell carcinoma[1][2][6].
