Overview

CRD5500 is a first-in-class, small molecule agonist of the Stimulator of Interferon Genes (STING) protein, developed by Curadev Pharma. Unlike natural cyclic dinucleotide ligands, CRD5500 binds to a unique allosteric site on STING located outside the dimeric cleft. This binding triggers the production of Type I interferons (IFN), which enhances T-cell infiltration into "cold" or non-inflamed tumors, leading to significant anti-tumor immune responses. Preclinical studies in human STING knock-in mice demonstrated efficacy across various models, including melanoma, pancreatic cancer, acute myeloid leukemia, and lung cancer. It is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors.
