Overview

Cre mRNA is a messenger RNA molecule that encodes the Cre recombinase protein, a site-specific DNA recombinase originally derived from the P1 bacteriophage. In the field of genetic engineering and drug delivery, Cre mRNA is a standard tool used to evaluate the efficiency and specificity of delivery vehicles, such as lipid nanoparticles (LNPs). When delivered into cells, the mRNA is translated into Cre recombinase, which then catalyzes the recombination of DNA between specific recognition sites known as loxP sites. This "Cre-loxP" system is extensively utilized in transgenic reporter mouse models (e.g., Ai14, Ai9, mTmG) to provide a permanent fluorescent readout of successful delivery and functional protein expression. Beyond its role as a benchmarking tool, Cre mRNA is employed to generate conditional knockout or knock-in animal models, enabling the study of gene function and the creation of complex disease models, such as lung cancer or neurodevelopmental disorders, in a spatially and temporally controlled manner.
