Creatine leucinate is a dietary supplement compound consisting of the nitrogenous organic acid creatine bonded to the branched-chain amino acid L-leucine. Developed primarily for the sports nutrition market, it is designed to enhance the ergogenic effects of standard creatine monohydrate by improving solubility and leveraging the anabolic signaling properties of leucine. The mechanism of action involves the delivery of creatine to myocytes via the creatine transporter (SLC6A8), where it is phosphorylated to phosphocreatine to facilitate rapid ATP regeneration during high-intensity anaerobic exercise. Simultaneously, the leucine component acts as a potent nutritional signal that activates the mechanistic target of rapamycin (mTOR) pathway, specifically the mTORC1 complex, to stimulate muscle protein synthesis and inhibit protein degradation.
