Overview

CREKA-GK8-QC is a smart, dual-targeted near-infrared (NIR-I) molecular imaging probe designed for the precise diagnosis and intraoperative fluorescence image-guided surgery of breast cancer. It combines a fibronectin-targeting peptide (CREKA) with an MMP-9-cleavable peptide sequence (GK8) and pairs them with a fluorescent dye (Cy5.5) and a fluorescence quencher (QSY21). This composition enables the probe to bind selectively to fibronectin in the tumor microenvironment and exhibit MMP-9-specific activatable fluorescence. The probe self-assembles into nanoparticles (~22 nm diameter), remaining quenched until encountering MMP-9, which cleaves the peptide and restores fluorescence, effectively highlighting tumors and micro-metastases for enhanced surgical guidance. Preclinical studies indicate strong tumor targeting, high imaging contrast, and negligible cytotoxicity[1][3].
