Overview

Crenolanib + cytarabine + idarubicin is a combination regimen consisting of three drugs: crenolanib, a second-generation, small molecule, oral tyrosine kinase inhibitor targeting FLT3 (Fms-like tyrosine kinase 3) and PDGFR (platelet-derived growth factor receptor), plus two classical cytotoxic chemotherapeutic agents, cytarabine (an antimetabolite) and idarubicin (an anthracycline DNA intercalator). This regimen is investigated for the treatment of adult patients with newly diagnosed or relapsed FLT3-mutant acute myeloid leukemia (AML), typically as an induction and consolidation regimen. Crenolanib is given sequentially after classic induction chemotherapy with high-dose cytarabine and idarubicin, with the strategy of maximizing initial cytotoxic effect and then targeting residual FLT3-mutant leukemic cells[1][3][5][7]. The combination aims to improve remission rates, depth of remission (MRD-negativity), and survival compared to chemotherapy alone. The regimen is currently investigational and not approved for standard clinical use.
