CRG-003

CRG-003 is a multiplex gene-edited, immune-evasive allogeneic CD19-targeting CAR-T therapy developed by Celregen Therapeutics for the treatment of B-cell-mediated autoimmune diseases. Utilizing CRISPR/Cas12 technology, the therapy undergoes precise knockouts of the TRAC and HLA-A/B genes to prevent graft-versus-host disease and host-mediated rejection, while selectively retaining HLA-C and overexpressing an immune evasion receptor to enhance persistence and minimize NK cell-mediated cytotoxicity. CRG-003 features a humanized CD19 binder with potency comparable to the FMC63 benchmark. It is currently being evaluated in a basket clinical trial for indications including systemic lupus erythematosus (SLE), idiopathic inflammatory myopathies (IIM), and ANCA-associated vasculitis (AAV).
