cRGDY-PEG-Cy5.5-C dots are ultrasmall core-shell silica nanoparticles designed for molecular imaging and image-guided surgery in oncology applications, particularly for sentinel lymph node (SLN) mapping and tumor localization[2][3]. These nanoparticles are functionalized with cyclic arginine-glycine-aspartic acid-tyrosine (cRGDY) peptides that selectively bind to alphaVbeta3 integrin receptors overexpressed on tumor cells[1][3]. The particles are coated with polyethylene glycol (PEG) to reduce non-specific uptake and encapsulate the near-infrared fluorophore Cy5.5 for enhanced optical imaging properties[3][2]. Upon intradermal administration, they enable real-time intraoperative visualization of cancerous tissues or metastatic nodes using fluorescence imaging systems[2][6]. Developed as a dual-modality platform, these particles can also be radiolabeled for PET imaging but the Cy5.5 version is primarily used for optical guidance.
