Overview

Crifortinib + azacitidine is an investigational drug combination comprising crifortinib, a small molecule tyrosine kinase inhibitor, and azacitidine, a DNA hypomethylating agent. Crifortinib targets and inhibits FLT3 (FMS-like tyrosine kinase 3), a receptor tyrosine kinase that is commonly mutated in acute myeloid leukemia (AML), thereby interfering with key signaling pathways essential for leukemia cell proliferation and survival. Azacitidine incorporates into DNA and RNA, inhibiting DNA methylation and resulting in the reactivation of silenced tumor suppressor genes and cell death. The combination is being explored (by analogy to other FLT3 inhibitors + azacitidine, such as quizartinib + azacitidine) for the treatment of AML, particularly in patients with FLT3 mutations or those who are unfit for intensive chemotherapy.
