Overview

Crisaborole is a non-steroidal, topical small molecule drug used for the treatment of mild to moderate atopic dermatitis (eczema) in adults and children. It acts as a phosphodiesterase 4 (PDE4) inhibitor, primarily targeting the PDE4B isoform, which plays a key role in regulating inflammatory cytokine production. By inhibiting PDE4, crisaborole increases intracellular cyclic adenosine monophosphate (cAMP), leading to suppression of pro-inflammatory mediators such as tumor necrosis factor alpha (TNFα) and various interleukins involved in immune-mediated skin diseases. This mechanism helps reduce inflammation, redness, itching, and rashes associated with eczema[2][3][5]. Crisaborole was originally developed by Anacor Pharmaceuticals and is marketed under the brand name Eucrisa.
