Crisantaspase (Erwinaze) is an L-asparaginase enzyme derived from the bacterium *Erwinia chrysanthemi*. It is primarily indicated for the treatment of acute lymphoblastic leukemia (ALL) and lymphoblastic lymphoma (LBL) in patients who have developed hypersensitivity to *Escherichia coli*-derived asparaginase. The drug's mechanism of action involves the enzymatic cleavage of the amino acid L-asparagine into L-aspartic acid and ammonia. Unlike normal cells, certain leukemic lymphoblasts lack the enzyme asparagine synthetase and are unable to synthesize L-asparagine de novo, making them dependent on exogenous sources for protein synthesis. Depletion of systemic L-asparagine by crisantaspase leads to the inhibition of protein synthesis and subsequent apoptosis of the malignant cells. Developed and marketed by Jazz Pharmaceuticals, Erwinaze is typically administered via intramuscular or intravenous injection as part of a multi-agent chemotherapeutic regimen.
